چکیده

The role of oxygen derived free radicals in gastric lesions induced by haemorrhagic shock and the protective effect of oxygen radical scavengers, allopurinol and ranitidine, were investigated. Forty five rabbits underwent haemorrhagic shock for 30 minutes and reinfusion of shed blood. They were killed 30 minutes later. The animals were divided in five groups: A (n = 10): Control, B (n = 10): intravenous ranitidine pretreatment, C (n = 10): oral allopurinol, 24 and 2 h before surgery; D (n = 10): intravenous pretreatment with superoxide Dismutase plus catalase, E (n = 5): 60 minute haemorrhagic shock without reinfusion and treatment. Erosions and/or petechiae in all animals in Group A were observed. Three animals in group B and C and 2 in group D (p less than 0.005, p less than 0.001) had gastric lesions. The lesions in the pretreatment groups were significantly smaller than in controls. Oxygen radicals plus HCl play an important role in shock induced gastric lesions. Oxygen radical antagonists show a significant protective role.

Oxygen-derived free radical scavengers: a new approach to the problem of refractory peptic ulceration.

Refractory peptic ulceration refers to ulcers which are slow to heal despite active treatment for at least three months. Oxygen-derived free radicals are cytotoxic and promote tissue injury. Twelve consecutive patients with refractory peptic ulceration (eight with duodenal ulcers and four with solitary pre-pyloric gastric ulcers) were treated using the radical scavengers allopurinol or dimethyl...

Free radicals and the pathogenesis of disease.

In this issue of the Medical Journal of Malaysia, A. S. Salim reports on his experience with free radical scavengers in the treatment of refractory peptic ulceration!. Although the number of patients in his study was small and the trial was open and non-comparative, the results obtained are interesting enough to justifY further research into the role of free radicals in the pathogenesis of pept...

The Protective Effect of Nortriptyline Against Gastric Lesions Induced by Indomethacin and Cold-shock Stress in Rats

Background: Gastric ulcer is among the most serious stomach disorder universally. Several effective drugs are employed in the management of this disease, although there have been adverse effects in some cases. The aim of this study was to investigate the effect of nortriptyline to protect against gastric lesions, induced by indomethacin or cold-stress in rats. Methods: Gastric lesions were ind...

Protective role of ascorbic acid in hemorrhage-induced cardiovascular depression.

Haemorrhagic shock is associated with hyperproduction of free radicals. Hence ascorbic acid (AA), a free radical scavenger, may be of benefit in such cases. We investigated in Wistar rats the role of free radicals in causing cardiovascular depression and the protective role of ascorbic acid, if any, in acute haemorrhage. The rats were catheterized for recording hemodynamic parameters. Acute hae...

Antioxidant vitamins and enzymatic and synthetic oxygen-derived free radical scavengers in the prevention and treatment of cardiovascular disease.

Oxygen-derived free radical formation can lead to cellular injury and death. Under normal situations, the human body has a free radical scavenger system (catalase, superoxide dismutase) that can detoxify free radicals. Antioxidant vitamins and enzymatic and synthetic oxygen-derived free radical scavengers have been used clinically to prevent the formation of oxidized LDL and to prevent reperfus...
